Collimated light passes through a lens and prism toward a thin sample layer under a cover plate; at the sample–prism interface the critical angle (θc) governs total internal reflection (TIR), producing a shadow line viewed through the eyepiece and scale. As an optical instrument within an analytical laboratory system, it measures refractive index (n) to estimate solute concentration, applied in biochemistry and clinical pathology for assessments such as urine specific gravity or serum total protein.
