Schematic of a Toll-like receptor (TLR), a single-pass transmembrane pattern-recognition receptor of the innate immune system. The horseshoe-shaped extracellular domain represents leucine-rich repeats that bind pathogen-associated molecular patterns (PAMPs), linked through a membrane-spanning segment to a cytosolic signaling domain (TIR, Toll/IL-1 receptor). In hierarchy, it is a membrane protein embedded in the cell or endosomal membrane of immune cells within tissues. In immunology and cell biology, it initiates signaling that activates NF-κB (nuclear factor kappa B) and induces cytokine production.
