We want to empower scientists to enable a more more visual and reachable science.<\/p>\n<p style=\"text-align: justify;\">In the post, I will show you a little of the original research&nbsp;by&nbsp;<a href=\"https:\/\/www.researchgate.net\/profile\/Yelena_Sterlin2\" target=\"_blank\" rel=\"noopener\">Yelena Sterlin<\/a> and colleagues. She is a&nbsp;Ph.D. student in the&nbsp;Faculty of Agriculture, Food and Environment at the&nbsp;Hebrew University of Jerusalem.<\/p>\n<p style=\"text-align: justify;\">Her new paper&nbsp;<a href=\"https:\/\/www.researchgate.net\/publication\/330976296_Optimized_Small-Molecule_Pull-Downs_Define_MLBP1_as_an_Acyl-Lipid-Binding_Protein\" target=\"_blank\" rel=\"noopener\">Optimized Small\u2010Molecule Pull\u2010Downs Define MLBP1 as an Acyl\u2010Lipid\u2010Binding Protein<\/a><strong>&nbsp;<\/strong>brings light to the&nbsp;protein-metabolite interactions in plants, with focus on the abscisic acid (ABA) receptors.&nbsp;She is also a mind the graph subscriber, so she created a infographic in <a href=\"https:\/\/mindthegraph.com\">mind the graph<\/a> to explain their methodology.<\/p>\n<h2 style=\"text-align: center;\"><strong>The importance of&nbsp;START domain-related proteins<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">According to <a href=\"https:\/\/www.nature.com\/articles\/s41598-018-22538-9\" target=\"_blank\" rel=\"noopener\">Nature Magazine<\/a>, abscisic acid (ABA) is the main phytohormone involved in abiotic stress response and its adaptation. Also, it is a candidate agrichemical.&nbsp; The ABA&nbsp;was first discovered in the 1960s. After that, it has since been found to be a key regulator of such diverse processes as dormancy, germination, seed development, and responses to biotic and abiotic stress.<\/p>\n<p style=\"text-align: justify;\">The earliest events involving the ABA signaling pathway involves ABA binding to&nbsp;PYR\/PYL\/RCARs receptors. The receptors belong to the START-domain superfamily.<\/p>\n<p style=\"text-align: justify;\">The START protein domains&nbsp;are a lipid-binding domain present in genomes of bacteria, protists, yeast, plants and animals. However, they are&nbsp;more common in plants. According to<a href=\"https:\/\/genomebiology.biomedcentral.com\/articles\/10.1186\/gb-2004-5-6-r41\" target=\"_blank\" rel=\"noopener\"> Schrick<\/a>, the presence of START domains in evolutionarily distant species such as animals and plants suggests a conserved mechanism for interaction of proteins with lipids\/sterols.<\/p>\n<p style=\"text-align: justify;\">The paper published&nbsp; in <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/30735592\" target=\"_blank\" rel=\"noopener\">The Plant Journal<\/a> focuses on&nbsp;PYR\/PYL\/RCARs ABA receptors.&nbsp; The&nbsp;researchers decided to optimize&nbsp;a method&nbsp;to allow identification of START&nbsp;domain and START domain-related protein ligands in plants, because<em>&nbsp;&#8220;the&nbsp;START domain protein ligands are indispensable to proper functioning of their protein&nbsp;both in animals and plants, and plant START domain proteins were demonstrated to be implicated in essential physiological processes&#8221;.<\/em><\/p>\n<h2 style=\"text-align: center;\"><strong>A visual Abstract of the methodology<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<h4 style=\"text-align: center;\"><strong>To explain their methodology, the scientists created an infographic with mind the graph illustrations. It&#8217;s eye-catching and easier to understand.&nbsp;<\/strong><\/h4>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">The infographic below shows the experimental workflow. As you can see,&nbsp;they expressed and purified four START proteins. They filtrated and incubated the supernatant containing native soluble fraction using streptavidin sepharose, &nbsp;an&nbsp;affinity&nbsp;resin for reliable and high<wbr>-resolution purification of biomolecules. Then, they analyzed aliquots&nbsp;using western blot. To identify the ligand, they&nbsp;eluted putative ligands and subjected to&nbsp; a LC -MS (liquid chromatography and mass spectrometry) system analysis. They adjusted the&nbsp;approach to discover START ligands.
