Vi vill ge forskare m\u00f6jlighet att skapa en mer visuell och l\u00e4ttillg\u00e4nglig vetenskap.<\/p>\n<p style=\"text-align: justify;\">I inl\u00e4gget kommer jag att visa dig lite av den ursprungliga forskningen av&nbsp;<a href=\"https:\/\/www.researchgate.net\/profile\/Yelena_Sterlin2\" target=\"_blank\" rel=\"noopener\">Jelena Sterlin<\/a> och medarbetare. Hon \u00e4r doktorand vid fakulteten f\u00f6r jordbruk, livsmedel och milj\u00f6 vid Hebrew University of Jerusalem.<\/p>\n<p style=\"text-align: justify;\">Hennes nya uppsats&nbsp;<a href=\"https:\/\/www.researchgate.net\/publication\/330976296_Optimized_Small-Molecule_Pull-Downs_Define_MLBP1_as_an_Acyl-Lipid-Binding_Protein\" target=\"_blank\" rel=\"noopener\">Optimerade sm\u00e5molekyl\u00e4ra pull-downs definierar MLBP1 som ett acyl-lipidbindande protein<\/a><strong>&nbsp;<\/strong>belyser protein-metabolit-interaktioner i v\u00e4xter, med fokus p\u00e5 abscisinsyra (ABA)-receptorer. Hon \u00e4r ocks\u00e5 en \"mind the graph\"-abonnent, s\u00e5 hon skapade en infografik i <a href=\"https:\/\/mindthegraph.com\">T\u00e4nk p\u00e5 grafen<\/a> att f\u00f6rklara sin metod.<\/p>\n<h2 style=\"text-align: center;\"><strong>Betydelsen av START-dom\u00e4nrelaterade proteiner<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">Enligt <a href=\"https:\/\/www.nature.com\/articles\/s41598-018-22538-9\" target=\"_blank\" rel=\"noopener\">Tidskriften Nature<\/a>Abscisinsyra (ABA) \u00e4r det viktigaste fytohormonet som \u00e4r involverat i abiotisk stressrespons och dess anpassning. Det \u00e4r ocks\u00e5 ett kandidat\u00e4mne f\u00f6r jordbrukskemikalier.  ABA uppt\u00e4cktes f\u00f6r f\u00f6rsta g\u00e5ngen p\u00e5 1960-talet. Sedan dess har det visat sig vara en viktig regulator f\u00f6r s\u00e5 olika processer som vilol\u00e4ge, groning, fr\u00f6utveckling och reaktioner p\u00e5 biotisk och abiotisk stress.<\/p>\n<p style=\"text-align: justify;\">De tidigaste h\u00e4ndelserna som involverar ABA-signalv\u00e4gen involverar ABA-bindning till PYR\/PYL\/RCARs-receptorer. Receptorerna tillh\u00f6r superfamiljen START-dom\u00e4ner.<\/p>\n<p style=\"text-align: justify;\">START-proteindom\u00e4nerna \u00e4r en lipidbindande dom\u00e4n som finns i arvsmassan hos bakterier, protister, j\u00e4st, v\u00e4xter och djur. De \u00e4r dock vanligare i v\u00e4xter. Enligt<a href=\"https:\/\/genomebiology.biomedcentral.com\/articles\/10.1186\/gb-2004-5-6-r41\" target=\"_blank\" rel=\"noopener\"> Schrick<\/a>F\u00f6rekomsten av START-dom\u00e4ner i evolution\u00e4rt avl\u00e4gsna arter som djur och v\u00e4xter tyder p\u00e5 en bevarad mekanism f\u00f6r interaktion mellan proteiner och lipider\/steroler.<\/p>\n<p style=\"text-align: justify;\">Artikeln publicerades i <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/30735592\" target=\"_blank\" rel=\"noopener\">Tidskriften f\u00f6r v\u00e4xter<\/a> fokuserar p\u00e5 PYR\/PYL\/RCARs ABA-receptorer.  Forskarna best\u00e4mde sig f\u00f6r att optimera en metod f\u00f6r identifiering av START-dom\u00e4ner och START-dom\u00e4nrelaterade proteinligander i v\u00e4xter, eftersom<em>&nbsp;\"START-dom\u00e4nproteinliganderna \u00e4r n\u00f6dv\u00e4ndiga f\u00f6r att deras protein skall fungera korrekt b\u00e5de hos djur och v\u00e4xter, och START-dom\u00e4nproteiner i v\u00e4xter har visat sig vara inblandade i viktiga fysiologiska processer\".<\/em><\/p>\n<h2 style=\"text-align: center;\"><strong>En visuell sammanfattning av metoden<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<h4 style=\"text-align: center;\"><strong>F\u00f6r att f\u00f6rklara sin metodik skapade forskarna en infografik med grafillustrationer. Det \u00e4r i\u00f6gonfallande och l\u00e4ttare att f\u00f6rst\u00e5.&nbsp;<\/strong><\/h4>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">Infografiken nedan visar det experimentella arbetsfl\u00f6det. Som du kan se uttryckte och renade de fyra START-proteinerna. De filtrerade och inkuberade supernatanten som inneh\u00f6ll den nativa l\u00f6sliga fraktionen med streptavidinsefaros, ett affinitetsharts f\u00f6r tillf\u00f6rlitlig och h\u00f6g<wbr>-uppl\u00f6sande rening av biomolekyler. D\u00e4refter analyserade de alikvoter med hj\u00e4lp av western blot. F\u00f6r att identifiera liganden eluerade de putativa ligander och utsattes f\u00f6r en LC -MS (v\u00e4tskekromatografi och masspektrometri) systemanalys. De anpassade metoden f\u00f6r att uppt\u00e4cka START-ligander.
