Det endocannabinoida systemet"},"content":{"rendered":"<p>Cannabisbehandlingar har f\u00e5tt stor uppm\u00e4rksamhet fr\u00e5n det vetenskapliga och medicinska samfundet \u00f6ver hela v\u00e4rlden under det senaste decenniet. <\/p>\n\n\n\n<p>Behandlingssvaren har visat sig vara mycket b\u00e4ttre j\u00e4mf\u00f6rt med konventionella l\u00e4kemedel. <\/p>\n\n\n\n<p>D\u00e4refter <strong>endocannabinoida systemet<\/strong> har uppm\u00e4rksammats i en exponentiell takt; forskare \u00e4r engagerade i att b\u00e4ttre f\u00f6rst\u00e5 hur detta system som finns i kroppen agerar och fungerar. <\/p>\n\n\n\n<p>Forskarna tittar d\u00e4rf\u00f6r n\u00e4rmare p\u00e5 detta f\u00f6r att kunna besvara l\u00e4karnas och patienternas fr\u00e5gor. <\/p>\n\n\n\n<p>Tidigare har vi till fullo f\u00f6rst\u00e5tt cannabisplantans terapeutiska effekter, men nu m\u00e5ste vetenskapen besvara fr\u00e5gor om <strong>endocannabinoida systemet<\/strong>. <\/p>\n\n\n\n<p>L\u00e5t oss se vad som redan \u00e4r k\u00e4nt.<\/p>\n\n\n\n<p>Den <strong>Endocannabinoida systemet (ES)<\/strong> \u00e4r en modulator av nervsystemet som fungerar som ett n\u00e4tverk i hela kroppen, en typ av regulator av det centrala nervsystemet. <\/p>\n\n\n\n<p>Hos vuxna har det endocannabinoida systemet ett stort inflytande p\u00e5 moduleringen av neuronal aktivitet - det p\u00e5verkar m\u00e5nga kognitiva och fysiologiska processer som hunger, s\u00f6mn, minne, \u00e5ngest, immunf\u00f6rsvar, motoriska f\u00e4rdigheter och andra. <\/p>\n\n\n\n<p>Den <strong>ES<\/strong> \u00e4r en lagspelare som inte fungerar som ett isolerat system utan \u00e4r kopplat till en m\u00e4ngd olika signaleringskaskader fr\u00e5n andra system i kroppen.<\/p>\n\n\n\n<p>Det finns tre huvudkomponenter som bildar ES - den <strong>cannabinoidreceptorer<\/strong>, <strong>endogena cannabinoider<\/strong>och <strong>enzymer<\/strong> ansvarar f\u00f6r syntes och nedbrytning av endocannabinoida molekyler. <\/p>\n\n\n\n<p>Den <strong>endogena cannabinoider \u00e4r<\/strong> en typ av molekyler som produceras av v\u00e5r kropp, och den finns ocks\u00e5 i naturen d\u00e4r cannabisplantan \u00e4r ett exempel. <\/p>\n\n\n\n<p>Det finns tre olika molekyler som vetenskapen idag vet kan trigga cannabinoidreceptorer - 2-AG och anandamid (AEA), som \u00e4r kortnamnen f\u00f6r 2-Arachidonoylglycerol respektive N-arachidonoyletanolamin, klassificeras b\u00e5da som endogena cannabinoider med lipidliknande strukturer. <\/p>\n\n\n\n<p>Den tredje, som kallas N-Arachidonoyl dopamine eller bara NADA, \u00e4r ocks\u00e5 en lipidstruktur och interagerar ocks\u00e5 med cannabinoidreceptorer<strong>.<\/strong> <\/p>\n\n\n\n<p>De tv\u00e5 f\u00f6rsta kan binda och aktivera flera signalv\u00e4gar f\u00f6r G-proteinkopplade receptorer, jonkanaler och k\u00e4rnreceptorer - medan den sista ansvarar f\u00f6r att detektera steroid- och sk\u00f6ldk\u00f6rtelhormoner. <\/p>\n\n\n\n<p>F\u00f6rutom likheterna i molekylstruktur, <strong>2-AG <\/strong>och <strong>anandamid<\/strong> spelar olika roller i <strong>ES,<\/strong> ha distinkta biokemiska cykler i kroppen - i missbruk<a href=\"https:\/\/www.ingentaconnect.com\/content\/ben\/cpd\/2012\/00000018\/00000032\/art00029\"> en granskad artikel publicerades 2012 om anandamids roll vid schizofreni<\/a>.\u00a0<\/p>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"aligncenter size-large\"><a href=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endogenous-cannabinoids.png\"><img decoding=\"async\" loading=\"lazy\" width=\"1024\" height=\"1024\" src=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endogenous-cannabinoids-1024x1024.png\" alt=\"\" class=\"wp-image-12137\" srcset=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endogenous-cannabinoids-1024x1024.png 1024w, https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endogenous-cannabinoids-300x300.png 300w, https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endogenous-cannabinoids-150x150.png 150w, https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endogenous-cannabinoids-768x768.png 768w, https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endogenous-cannabinoids-1536x1536.png 1536w, https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endogenous-cannabinoids.png 1600w\" sizes=\"(max-width: 1024px) 100vw, 1024px\" \/><\/a><\/figure><\/div>\n\n\n\n<p>Flera studier har visat att endogena cannabinoider syntetiseras av kroppen efter behov, med andra ord \"p\u00e5 beg\u00e4ran\". <\/p>\n\n\n\n<p>Detta inneb\u00e4r att vi inte syntetiserar endogena cannabinoider hela tiden, de flyter inte runt i blodomloppet och v\u00e4ntar p\u00e5 att bli kallade, de syntetiseras p\u00e5 en specifik plats n\u00e4r de beh\u00f6vs.<\/p>\n\n\n\n<p>De tv\u00e5 <strong>cannabinoider receptorer<\/strong> k\u00e4nd idag,<strong> CB1<\/strong> och <strong>CB2<\/strong> \u00e4r b\u00e5da G-proteinkopplade receptorer. <\/p>\n\n\n\n<p>Receptorerna har olika funktioner, de kan interagera med olika molekyler och aktivera olika signalv\u00e4gar, och detta kan leda till specifika eller divergerande biologiska effekter.<\/p>\n\n\n\n<p>De flesta CB1-receptorer finns i det centrala nervsystemet, men de kan ocks\u00e5 finnas i organ som lever, hud och fettv\u00e4vnad. <\/p>\n\n\n\n<p>I nervsystemet finns CB1 i flera synaptiska terminaler och har en viktig roll i moduleringen av den synaptiska transmissionen. <\/p>\n\n\n\n<p>Denna receptor har ocks\u00e5 en f\u00f6rm\u00e5ga att heterodimerisera - binda till olika strukturer av sig sj\u00e4lv - med andra G-proteinkopplade receptorer, som dopamin D2, hypokretin och opioidreceptorer. Faktum \u00e4r att ett f\u00e5tal studier visade<a href=\"https:\/\/pubmed.ncbi.nlm.nih.gov\/14706563\/\"> att cannabis hj\u00e4lper till med sm\u00e4rtlindring och minskar doserna av opioider.<\/a>\u00a0<\/p>\n\n\n\n<p>CB2-receptorerna \u00e4r rikligt f\u00f6rekommande i immunsystemets celler, och trots att de uttrycks<s> <\/s>I mycket l\u00e4gre niv\u00e5er \u00e4n CB1 finns \u00e4ven CB2 i nervceller. <\/p>\n\n\n\n<p>Denna receptor finns i olika organ som levern, mj\u00e4lten, bukspottk\u00f6rteln, tjocktarmen och skelettet. <\/p>\n\n\n\n<p>Forskare beskriver CB2-receptoraktivering som en antiinflammatorisk faktor i inflammationsscenarier. CB1 \u00e4r den huvudsakliga receptorn f\u00f6r anandamid och THC. <\/p>\n\n\n\n<p>B\u00e5de anandamid och THC klassificeras som partiella CB1-receptoragonister, vilket inneb\u00e4r att molekylen har mycket l\u00e5g effekt n\u00e4r det g\u00e4ller att framkalla en receptorrespons, vilket direkt p\u00e5verkar de terapeutiska effekterna. <\/p>\n\n\n\n<p>Samtidigt klassificeras 2-AG - som finns i st\u00f6rre m\u00e4ngd i hj\u00e4rnan \u00e4n anandamid - som en h\u00f6geffektiv agonist f\u00f6r b\u00e5da receptorerna, CB1 och CB2, med kapacitet att binda till receptorn och aktivera och producera en biologisk respons p\u00e5 h\u00f6g niv\u00e5. <\/p>\n\n\n\n<p>Under tiden klassificeras CBD som en antagonist f\u00f6r b\u00e5da receptorerna, vilket inneb\u00e4r att molekylen ansluter sig till receptorn utan att aktivera den, vilket f\u00f6rhindrar att andra molekyler ansluter och aktiverar receptorn.<\/p>\n\n\n\n<div class=\"wp-block-image\"><figure class=\"aligncenter size-large\"><a href=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endocannabinoid-system.png\"><img decoding=\"async\" loading=\"lazy\" width=\"471\" height=\"666\" src=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endocannabinoid-system.png\" alt=\"\" class=\"wp-image-12138\" srcset=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endocannabinoid-system.png 471w, https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/01\/endocannabinoid-system-212x300.png 212w\" sizes=\"(max-width: 471px) 100vw, 471px\" \/><\/a><\/figure><\/div>\n\n\n\n<p>P\u00e5 grund av det stora antalet endocannabinoidreceptorer i hj\u00e4rnan har ett f\u00e5tal psykiska sjukdomar f\u00f6rknippats med dysregleringar av endocannabinoidsystemet och genetiska polymorfismer, t.ex. psykos, schizofreni, \u00e5ngest, depression och autism. <\/p>\n\n\n\n<p>Forskare har f\u00f6rs\u00f6kt f\u00f6rst\u00e5 och ta reda p\u00e5 det exakta sambandet mellan eventuella mutationer i det endocannabinoida systemet och dessa sjukdomar.<\/p>\n\n\n\n<p>Idag \u00e4r det m\u00f6jligt f\u00f6r patienter att drabbas av en<a href=\"https:\/\/www.iproprium.com\/mycannabis-code\/\"> f\u00f6retag att analysera deras genetiska kod<\/a> och f\u00e5 alla m\u00f6jliga mutationer av deras endocannabinoida systemkod och f\u00f6rst\u00e5 hur deras kropp hanterar cannabismolekyler som THC, CBD och m\u00e5nga andra.&nbsp;&nbsp;<\/p>\n\n\n\n<p>Det finns idag inte mycket kunskap om den allosteriska moduleringen av CB2-receptorn, men f\u00f6r CB1 finns det flera k\u00e4nda allosteriska modulatorer. <\/p>\n\n\n\n<p>Allosteriska modulatorer kan \u00e4ndra kinetiken och\/eller effektiviteten hos receptorn - \u00e4r en enzymatisk reglering som k\u00e4nnetecknas av att en effektormolekyl binds till en annan region \u00e4n enzymets aktiva site, denna sekund\u00e4ra site kallas ortostatisk site.<\/p>\n\n\n\n<p>Ett viktigt faktum att n\u00e4mna \u00e4r att CBD \u00e4r en negativ allosterisk modulator av CB1, vilket minskar CB1-aktiveringen av THC. <\/p>\n\n\n\n<p>Med andra ord har THC sina effekter minskat genom effekten av CBD.
