Zato vedno radi poka\u017eemo neverjetna dela na\u0161ih uporabnikov kot navdihujo\u010d primer vizualne abstrakcije. Znanstvenike \u017eelimo opolnomo\u010diti, da bi omogo\u010dili bolj vizualno in dosegljivo znanost.<\/p>\n<p style=\"text-align: justify;\">V prispevku vam bom predstavil del\u010dek izvirne raziskave, ki jo je&nbsp;<a href=\"https:\/\/www.researchgate.net\/profile\/Yelena_Sterlin2\" target=\"_blank\" rel=\"noopener\">Yelena Sterlin<\/a> in sodelavci. Je doktorska \u0161tudentka na Fakulteti za kmetijstvo, prehrano in okolje na Hebrejski univerzi v Jeruzalemu.<\/p>\n<p style=\"text-align: justify;\">Njen novi \u010dlanek&nbsp;<a href=\"https:\/\/www.researchgate.net\/publication\/330976296_Optimized_Small-Molecule_Pull-Downs_Define_MLBP1_as_an_Acyl-Lipid-Binding_Protein\" target=\"_blank\" rel=\"noopener\">Optimizirani izvle\u010dki majhnih molekul opredeljujejo MLBP1 kot beljakovino, ki ve\u017ee acil-lipide<\/a><strong>&nbsp;<\/strong>osvetljuje interakcije med proteini in metaboliti v rastlinah, s poudarkom na receptorjih za abscisi\u010dno kislino (ABA). Je tudi naro\u010dnica programa mind the graph, zato je ustvarila infografiko v <a href=\"https:\/\/mindthegraph.com\">upo\u0161tevajte graf<\/a> pojasniti svojo metodologijo.<\/p>\n<h2 style=\"text-align: center;\"><strong>Pomen proteinov, povezanih z domeno START<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">Po podatkih <a href=\"https:\/\/www.nature.com\/articles\/s41598-018-22538-9\" target=\"_blank\" rel=\"noopener\">Revija Nature<\/a>abscisi\u010dna kislina (ABA) je glavni fitohormon, ki sodeluje pri odzivanju na abiotski stres in prilagajanju nanj. Prav tako je kandidat za agrokemikalije.  ABA je bila prvi\u010d odkrita v \u0161estdesetih letih prej\u0161njega stoletja. Nato je bilo ugotovljeno, da je klju\u010dni regulator razli\u010dnih procesov, kot so mirovanje, kalitev, razvoj semen ter odzivi na biotski in abiotski stres.<\/p>\n<p style=\"text-align: justify;\">Najzgodnej\u0161i dogodki, ki vklju\u010dujejo signalno pot ABA, vklju\u010dujejo vezavo ABA na receptorje PYR\/PYL\/RCARs. Receptorji spadajo v naddru\u017eino z domeno START.<\/p>\n<p style=\"text-align: justify;\">Proteinske domene START so domene, ki ve\u017eejo lipide in so prisotne v genomih bakterij, protistov, kvasovk, rastlin in \u017eivali. Vendar so pogostej\u0161e pri rastlinah. Po podatkih<a href=\"https:\/\/genomebiology.biomedcentral.com\/articles\/10.1186\/gb-2004-5-6-r41\" target=\"_blank\" rel=\"noopener\"> Schrick<\/a>, prisotnost domen START pri evolucijsko oddaljenih vrstah, kot so \u017eivali in rastline, ka\u017ee na ohranjen mehanizem interakcije proteinov z lipidi\/steroli.<\/p>\n<p style=\"text-align: justify;\">Dokument, objavljen v <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/30735592\" target=\"_blank\" rel=\"noopener\">\u010casopis za rastline<\/a> se osredoto\u010da na receptorje ABA PYR\/PYL\/RCAR.  Raziskovalci so se odlo\u010dili optimizirati metodo, ki bi omogo\u010dila identifikacijo ligandov domene START in z domeno START povezanih proteinov v rastlinah, ker<em>&nbsp;\"ligandi proteinov z domeno START so nepogre\u0161ljivi za pravilno delovanje njihovih proteinov tako pri \u017eivalih kot pri rastlinah, za rastlinske proteine z domeno START pa je bilo dokazano, da so vpleteni v bistvene fiziolo\u0161ke procese\".<\/em><\/p>\n<h2 style=\"text-align: center;\"><strong>Vizualni povzetek metodologije<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<h4 style=\"text-align: center;\"><strong>Da bi pojasnili svojo metodologijo, so znanstveniki pripravili infografiko z ilustracijami grafov. Ta je privla\u010dna za o\u010di in la\u017eje razumljiva.&nbsp;<\/strong><\/h4>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">Spodnja infografika prikazuje eksperimentalni potek dela. Kot lahko vidite, so izrazili in o\u010distili \u0161tiri beljakovine START. Supernatant, ki je vseboval nativno topno frakcijo, so filtrirali in inkubirali s streptavidinsko sefarozo, afinitetno smolo za zanesljivo in visoko<wbr>-\u010di\u0161\u010denje biomolekul z visoko lo\u010dljivostjo. Nato so alikvote analizirali z metodo western blot. Za identifikacijo liganda so eluirali domnevne ligande in jih analizirali s sistemom LC-MS (teko\u010dinska kromatografija in masna spektrometrija). Prilagodili so pristop za odkrivanje ligandov START. S prilagoditvijo analiti\u010dnih pogojev pa lahko metodo uporabimo za ligande razli\u010dne kemijske narave.<\/p>\n<p style=\"text-align: justify;\">\u010ce \u017eelite svoje raziskave prikazati z znanstvenimi ilustracijami in vizualnimi povzetki, lahko uporabite tudi Mind the Graph.
