ACE2 nima samo ene strukture, ampak ima lahko ve\u010d struktur med vrstami, pri \u010demer ohranja enako funkcijo. <\/p>\n\n\n\n<p>ACE2 je namre\u010d uvr\u0161\u010den med ortologne gene, torej gene, ki so prisotni pri razli\u010dnih vrstah in izvirajo iz skupnega gena prednikov.<\/p>\n\n\n<div class=\"wp-block-image\">\n<figure class=\"aligncenter size-large\"><a href=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes.png\"><img decoding=\"async\" loading=\"lazy\" width=\"433\" height=\"512\" src=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes.png\" alt=\"\" class=\"wp-image-12239\" srcset=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes.png 433w, https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes-254x300.png 254w\" sizes=\"(max-width: 433px) 100vw, 433px\" \/><\/a><\/figure><\/div>\n\n\n<p>Te strukturne razlike so o\u010ditno razlog, da se virus SARS-CoV-2 lahko ve\u017ee na ACE2 nekaterih vrst, drugih pa ne. <\/p>\n\n\n\n<p>Ko se virusni protein-\u0161pica pove\u017ee z ACE2, virus vstopi v celico in za\u010dne uporabljati gostiteljeve celi\u010dne mehanizme za razmno\u017eevanje, kar privede do oku\u017ebe.<\/p>\n\n\n\n<p>Bolj\u0161e razumevanje receptorja ACE2 bi tako zagotovo vodilo k natan\u010dnej\u0161i razlagi, zakaj so nekatere \u017eivalske vrste dovzetne za oku\u017ebe, druge pa ne. <\/p>\n\n\n\n<p>Poleg tega bi znanstvenikom pomagal tudi pri razvoju zdravil, saj bi uporabili protivirusne strategije, kot je izdelava la\u017enih omaric, na katere bi se virus vezal namesto na dejanske omarice gostitelja. <\/p>\n\n\n\n<p>ter za bolj\u0161e sledenje vmesnim \u017eivalim gostiteljicam za prihodnje oku\u017ebe z virusom.&nbsp;<\/p>\n\n\n\n<p>Zato je bil cilj projekta opisati tridimenzionalno strukturo ACE2 in njegove interakcije na atomski ravni z uporabo ra\u010dunalni\u0161kega modeliranja.<a href=\"https:\/\/journals.plos.org\/ploscompbiol\/article?id=10.1371\/journal.pcbi.1008449\"> Rodrigues in sodelavci<\/a> delo. <\/p>\n\n\n\n<p>Tovrstna \u0161tudija karakteristik je eksperimentalno draga in se ne uporablja v velikem obsegu, saj je z njo te\u017eko dose\u010di prepri\u010dljive podatke, medtem ko je ra\u010dunalni\u0161ko modeliranje hitro in poceni ter je zelo koristno za zapolnitev nekaterih vrzeli v znanju o strukturnih okvirih.<\/p>\n\n\n\n<p class=\"has-large-font-size\">V \u0161tudiji je bilo analiziranih 28 \u017eivalskih vrst, pri \u010demer je bil ustvarjen 3D-model ACE2, ki je bil povezan z virusnim proteinom spike. <\/p>\n\n\n\n<p>Rezultati so pokazali, da so v aminokislinskem zaporedju ACE2 klju\u010dna mesta, kjer se zaradi mutacij virusni protein ne more vezati na gostiteljsko celico. <\/p>\n\n\n\n<p>Natan\u010dneje, v zaporedju ACE2 manjka ena aminokislina, odgovorna za medmolekularni solni most med ACE2 in vezavnim mestom virusnega proteina, kot so poro\u010dali Rodrigues in njegova ekipa. <\/p>\n\n\n\n<p>Ta rezultat potrjujejo druge objavljene \u0161tudije, ki so eksperimentalno dokazale, da je pomanjkanje tega mutanta odsotnost negativno nabite aminokisline, ki omogo\u010da vezavo virusnega proteina.<\/p>\n\n\n\n<p class=\"has-large-font-size\">\u0160tudija tudi ka\u017ee, da je oku\u017eba z virusom SARS-CoV-2 bolj zapletena, kot se je domnevalo doslej, saj lahko gostitelja oku\u017ei na ve\u010d na\u010dinov. <\/p>\n\n\n\n<p>Zato je pomembno pojasniti, da lahko zmanj\u0161anje razpolo\u017eljivosti ACE2 zmanj\u0161a mo\u017enost oku\u017ebe, vendar pa ne prepre\u010di nastanka oku\u017ebe.<\/p>\n\n\n\n<p>Izra\u010dunskih modelov ni mogo\u010de uporabiti samo za napovedovanje tveganj oku\u017ebe vrst - za zanesljivej\u0161o ugotovitev so potrebni dodatni eksperimentalni podatki.<\/p>\n\n\n\n<p>Zato mislim, da moramo \u0161e naprej upo\u0161tevati pandemske protokole in delati to, kar smo \u017ee po\u010deli...
