Je doktorandkou na Fakulte po\u013enohospod\u00e1rstva, potravin\u00e1rstva a \u017eivotn\u00e9ho prostredia Hebrejskej univerzity v Jeruzaleme.<\/p>\n<p style=\"text-align: justify;\">Jej nov\u00fd dokument&nbsp;<a href=\"https:\/\/www.researchgate.net\/publication\/330976296_Optimized_Small-Molecule_Pull-Downs_Define_MLBP1_as_an_Acyl-Lipid-Binding_Protein\" target=\"_blank\" rel=\"noopener\">Optimalizovan\u00e9 Pull-Downs s mal\u00fdmi molekulami definuj\u00fa MLBP1 ako prote\u00edn via\u017euci acyl-lypidy<\/a><strong>&nbsp;<\/strong>vn\u00e1\u0161a svetlo do interakci\u00ed medzi prote\u00ednmi a metabolitmi v rastlin\u00e1ch so zameran\u00edm na receptory kyseliny abscisovej (ABA). Je tie\u017e predplatite\u013eom mind the graph, tak\u017ee vytvorila infografiku v <a href=\"https:\/\/mindthegraph.com\">Pozor na graf<\/a> vysvetli\u0165 svoju metodiku.<\/p>\n<h2 style=\"text-align: center;\"><strong>V\u00fdznam prote\u00ednov s\u00favisiacich s dom\u00e9nou START<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">Pod\u013ea <a href=\"https:\/\/www.nature.com\/articles\/s41598-018-22538-9\" target=\"_blank\" rel=\"noopener\">\u010casopis Nature<\/a>, kyselina abscisov\u00e1 (ABA) je hlavn\u00fdm fytohorm\u00f3nom, ktor\u00fd sa podie\u013ea na reakcii na abiotick\u00fd stres a na adapt\u00e1cii na\u0148. Je tie\u017e kandid\u00e1tom na agrochemik\u00e1lie.  ABA bola prv\u00fdkr\u00e1t objaven\u00e1 v 60. rokoch 20. storo\u010dia. Potom sa zistilo, \u017ee odvtedy je k\u013e\u00fa\u010dov\u00fdm regul\u00e1torom tak\u00fdch rozmanit\u00fdch procesov, ako je dormancia, kl\u00ed\u010denie, v\u00fdvoj semien a reakcie na biotick\u00fd a abiotick\u00fd stres.<\/p>\n<p style=\"text-align: justify;\">Najstar\u0161ie udalosti zah\u0155\u0148aj\u00face sign\u00e1lnu dr\u00e1hu ABA zah\u0155\u0148aj\u00fa v\u00e4zbu ABA na receptory PYR\/PYL\/RCARs. Tieto receptory patria do superrodiny s dom\u00e9nou START.<\/p>\n<p style=\"text-align: justify;\">Prote\u00ednov\u00e9 dom\u00e9ny START s\u00fa dom\u00e9ny via\u017euce lipidy, ktor\u00e9 sa nach\u00e1dzaj\u00fa v gen\u00f3moch bakt\u00e9ri\u00ed, protistov, kvasiniek, rastl\u00edn a \u017eivo\u010d\u00edchov. \u010castej\u0161ie sa v\u0161ak vyskytuj\u00fa v rastlin\u00e1ch. Pod\u013ea<a href=\"https:\/\/genomebiology.biomedcentral.com\/articles\/10.1186\/gb-2004-5-6-r41\" target=\"_blank\" rel=\"noopener\"> Schrick<\/a>, pr\u00edtomnos\u0165 dom\u00e9n START u evolu\u010dne vzdialen\u00fdch druhov, ako s\u00fa \u017eivo\u010d\u00edchy a rastliny, nazna\u010duje konzervovan\u00fd mechanizmus interakcie prote\u00ednov s lipidmi\/sterolmi.<\/p>\n<p style=\"text-align: justify;\">Dokument uverejnen\u00fd v \u010dasopise <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/30735592\" target=\"_blank\" rel=\"noopener\">Rastlinn\u00fd \u010dasopis<\/a> sa zameriava na receptory ABA PYR\/PYL\/RCAR.  V\u00fdskumn\u00edci sa rozhodli optimalizova\u0165 met\u00f3du, ktor\u00e1 by umo\u017enila identifikova\u0165 ligandy START dom\u00e9ny a prote\u00ednov pr\u00edbuzn\u00fdch START dom\u00e9ne v rastlin\u00e1ch, preto\u017ee<em>&nbsp;\"ligandy prote\u00ednov s dom\u00e9nou START s\u00fa nevyhnutn\u00e9 pre spr\u00e1vne fungovanie ich prote\u00ednov u \u017eivo\u010d\u00edchov aj rastl\u00edn a bolo preuk\u00e1zan\u00e9, \u017ee rastlinn\u00e9 prote\u00edny s dom\u00e9nou START sa podie\u013eaj\u00fa na z\u00e1kladn\u00fdch fyziologick\u00fdch procesoch\".<\/em><\/p>\n<h2 style=\"text-align: center;\"><strong>Vizu\u00e1lna Abstrakt metodiky<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<h4 style=\"text-align: center;\"><strong>Na vysvetlenie svojej metodiky vedci vytvorili infografiku s ilustr\u00e1ciami grafov. Je p\u00fatav\u00e1 a \u013eah\u0161ie pochopite\u013en\u00e1.&nbsp;<\/strong><\/h4>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">Na infografike ni\u017e\u0161ie je zn\u00e1zornen\u00fd experiment\u00e1lny pracovn\u00fd postup. Ako m\u00f4\u017eete vidie\u0165, exprimovali a purifikovali \u0161tyri prote\u00edny START. Supernatant obsahuj\u00faci nat\u00edvnu rozpustn\u00fa frakciu prefiltrovali a inkubovali pomocou streptavid\u00ednovej sefar\u00f3zy, afinitnej \u017eivice na spo\u013eahliv\u00e9 a vysoko<wbr>-purifik\u00e1cia biomolek\u00fal s vysok\u00fdm rozl\u00ed\u0161en\u00edm. Potom analyzovali alikvoty pomocou western blotu. Na identifik\u00e1ciu ligandu eluovali predpokladan\u00e9 ligandy a podrobili ich anal\u00fdze syst\u00e9mom LC -MS (kvapalinov\u00e1 chromatografia a hmotnostn\u00e1 spektrometria).
