O neoplasma de melan\u00f3citos na pele, mucosa ou uvea \u00e9 conhecido como melanoma. Apesar de sua representa\u00e7\u00e3o de 5% em todas as malignidades cut\u00e2neas, ele \u00e9 respons\u00e1vel por mais de 75% de todas as mortes por c\u00e2ncer de pele.\u00a0<\/p>\n\n\n\n<p>Embora a associa\u00e7\u00e3o positiva entre a exposi\u00e7\u00e3o UV e o desenvolvimento do melanoma seja bem conhecida, os mecanismos epigen\u00e9ticos subjacentes s\u00e3o em grande parte inexplorados nos tecidos do melanoma humano. Uma coorte multi-\u00e9tnica de 112 c\u00e9lulas de melanoma cut\u00e2neo foi inclu\u00edda neste estudo para perfil cl\u00ednico, epigenoma (metiloma de DNA), genoma e transcriptoma. Neste estudo, eles identificam mudan\u00e7as induzidas por raios UV em vias imunol\u00f3gicas e regulat\u00f3rias que podem ser propulsoras de c\u00e2ncer atrav\u00e9s de m\u00faltiplos MICs.<\/p>\n\n\n\n<p>Al\u00e9m de ser o gene mais cr\u00edtico, o TAPBP (Tap binding protein gene) est\u00e1 criticamente envolvido na fun\u00e7\u00e3o imunol\u00f3gica, com v\u00e1rios locais de metila\u00e7\u00e3o alterados por raios UV que foram validados por seq\u00fcenciamento direcionado, proporcionando oportunidades clinicamente aplic\u00e1veis. O MHC-I interage com o transportador associado ao processamento de ant\u00edgenos atrav\u00e9s do TAPBP, um membro da superfam\u00edlia da imunoglobulina. Uma s\u00e9rie de c\u00e2nceres tem mostrado desregulamenta\u00e7\u00e3o da prote\u00edna TAPBP (tapasina), que \u00e9 restaurada ap\u00f3s a exposi\u00e7\u00e3o a citocinas.\u00a0<\/p>\n\n\n\n<p>Isto sugere que a express\u00e3o deficiente do TAPBP pode ser causada por desregulamenta\u00e7\u00e3o e n\u00e3o por mudan\u00e7as estruturais.
