A ACE2 n\u00e3o tem apenas uma \u00fanica estrutura, ela pode ter m\u00faltiplas estruturas de esp\u00e9cies cruzadas, mantendo a mesma fun\u00e7\u00e3o. <\/p>\n\n\n\n<p>Na verdade, o ACE2 \u00e9 classificado como um gene ortol\u00f3gico, ou seja, um gene presente em diferentes esp\u00e9cies que se originou de um gene ancestral comum.<\/p>\n\n\n<div class=\"wp-block-image\">\n<figure class=\"aligncenter size-large\"><a href=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes.png\"><img decoding=\"async\" loading=\"lazy\" width=\"433\" height=\"512\" src=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes.png\" alt=\"\" class=\"wp-image-12239\" srcset=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes.png 433w, https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes-254x300.png 254w\" sizes=\"(max-width: 433px) 100vw, 433px\" \/><\/a><\/figure><\/div>\n\n\n<p>Estas diferen\u00e7as estruturais s\u00e3o aparentemente a raz\u00e3o pela qual o v\u00edrus SARS-CoV-2 pode se ligar \u00e0 ACE2 de algumas esp\u00e9cies e n\u00e3o a outras. <\/p>\n\n\n\n<p>Uma vez que a prote\u00edna-spike viral \u00e9 conectada ao ACE2, o v\u00edrus entra na c\u00e9lula e come\u00e7a a usar o maquin\u00e1rio celular hospedeiro para se multiplicar levando \u00e0 infec\u00e7\u00e3o.<\/p>\n\n\n\n<p>Como resultado, ter um melhor entendimento sobre o receptor ACE2 certamente levaria a uma explica\u00e7\u00e3o mais precisa do porqu\u00ea de algumas esp\u00e9cies de animais serem suscet\u00edveis \u00e0 infec\u00e7\u00e3o enquanto outras n\u00e3o o s\u00e3o. <\/p>\n\n\n\n<p>Al\u00e9m disso, ajudaria os cientistas no desenvolvimento de medicamentos tamb\u00e9m, aplicando estrat\u00e9gias antivirais, como a produ\u00e7\u00e3o de arm\u00e1rios falsos para que o v\u00edrus se ligue em vez do arm\u00e1rio real do hospedeiro. <\/p>\n\n\n\n<p>Assim como para melhor rastrear os animais hospedeiros intermedi\u00e1rios para futuras infec\u00e7\u00f5es por v\u00edrus.&nbsp;<\/p>\n\n\n\n<p>Assim, caracterizar a estrutura tridimensional da ACE2 e suas intera\u00e7\u00f5es a n\u00edvel at\u00f4mico utilizando modelagem computacional, foi o objetivo de<a href=\"https:\/\/journals.plos.org\/ploscompbiol\/article?id=10.1371\/journal.pcbi.1008449\"> Rodrigues e colegas<\/a> trabalho. <\/p>\n\n\n\n<p>Este tipo de estudo de caracteriza\u00e7\u00e3o \u00e9 experimentalmente caro e n\u00e3o aplic\u00e1vel em larga escala, tornando dif\u00edcil chegar a dados conclusivos, enquanto isso, a modelagem computacional \u00e9 r\u00e1pida e barata, sendo muito ben\u00e9fica para preencher algumas lacunas no conhecimento estrutural da estrutura.<\/p>\n\n\n\n<p class=\"has-large-font-size\">O estudo, levou 28 esp\u00e9cies animais para an\u00e1lise, gerando um modelo 3D da ACE2 colada \u00e0 prote\u00edna do espig\u00e3o viral. <\/p>\n\n\n\n<p>Os resultados mostraram que existem locais-chave na sequ\u00eancia de amino\u00e1cidos ACE2, onde, devido a muta\u00e7\u00f5es, a prote\u00edna viral n\u00e3o podia se ligar \u00e0 c\u00e9lula hospedeira. <\/p>\n\n\n\n<p>Especificamente, \u00e0 seq\u00fc\u00eancia ACE2 falta um amino\u00e1cido respons\u00e1vel por uma ponte intermolecular salina observada entre ACE2 e o local de liga\u00e7\u00e3o da prote\u00edna viral, como relatado por Rodrigues e sua equipe. <\/p>\n\n\n\n<p>Este resultado \u00e9 apoiado por outros estudos publicados, que demonstraram experimentalmente que esta falta mutante \u00e9 a aus\u00eancia de amino\u00e1cido carregado negativamente, permitindo a liga\u00e7\u00e3o da prote\u00edna viral.<\/p>\n\n\n\n<p class=\"has-large-font-size\">O estudo tamb\u00e9m indica que a infec\u00e7\u00e3o pelo SARS-CoV-2 \u00e9 mais complexa do que se pensava anteriormente, com mais de uma maneira de infectar o hospedeiro. <\/p>\n\n\n\n<p>Assim, \u00e9 importante esclarecer que a redu\u00e7\u00e3o da disponibilidade do ACE2 pode diminuir as chances de infec\u00e7\u00e3o; no entanto, isso n\u00e3o impede que a infec\u00e7\u00e3o ocorra.<\/p>\n\n\n\n<p>E que os modelos de computa\u00e7\u00e3o n\u00e3o podem ser usados sozinhos para prever os riscos de infec\u00e7\u00e3o das esp\u00e9cies - dados experimentais adicionais s\u00e3o necess\u00e1rios para se ter uma conclus\u00e3o mais confi\u00e1vel.<\/p>\n\n\n\n<p>Portanto, acho que devemos continuar seguindo os protocolos pand\u00eamicos e fazer o que j\u00e1 estamos fazendo...
