De na\u00efeve ongesorteerde cellen werden gekweekt in FGF2-bevattend CSC-medium of met de FGFR-remmer SU-5402.<\/p>\n\n\n\n<h2><strong>De rol van fibroblast groeifactor 2 (FGF2) in de zelfvernieuwing van colorectale kankerstamcellen is onderzocht<\/strong><\/h2>\n\n\n\n<p>In 'FGF Signalling in the Self-Renewal of Colon Cancer Organoids', meldden J\u00f6rg Otte en collega's (2019) dat wanneer na\u00efeve enkelvoudige cellen werden ingebed in Matrigel en gekweekt in het CSC-medium, een meer complexe zelforganisatie werd waargenomen in alle geanalyseerde pati\u00ebnten. <\/p>\n\n\n\n<p>Genen die rechtstreeks verband houden met EMT in combinatie met een mesenchymaal en zelfvernieuwend fenotype (TGFB3, ID1, ID2 en ID4) bleken overge\u00ebxpresseerd te zijn in organo\u00efden. <\/p>\n\n\n\n<p>Terwijl FGFR-remming leidde tot een versterkte MAPK-signalering, constateerden zij ge\u00efnduceerde differentiatie, een verlies van de meeste stamcelmarkers en een epitheliaal fenotype. <\/p>\n\n\n\n<p>Kankerstamcellen zijn een subpopulatie van kwaadaardige cellen die zichzelf kunnen vernieuwen en kunnen dienen als een permanente bron voor gedifferentieerde tumorcellen. De GO-termen \"negatieve regulering van BMP-signaalroute\" en \"SMAD-eiwitsignaaltransductie\" werden gedetecteerd in onze hESC-kweek,\" stelde Otte.<\/p>\n\n\n\n<p>De auteurs geven toe dat \"GO-term-analyse van deze genenset slechts enkele significante resultaten opleverde vanwege het geringe aantal gemeenschappelijk gereguleerde genen.
