Elle est \u00e9tudiante en doctorat \u00e0 la facult\u00e9 d'agriculture, d'alimentation et d'environnement de l'universit\u00e9 h\u00e9bra\u00efque de J\u00e9rusalem.<\/p>\n<p style=\"text-align: justify;\">Son nouveau document&nbsp;<a href=\"https:\/\/www.researchgate.net\/publication\/330976296_Optimized_Small-Molecule_Pull-Downs_Define_MLBP1_as_an_Acyl-Lipid-Binding_Protein\" target=\"_blank\" rel=\"noopener\">Des analyses optimis\u00e9es de petites mol\u00e9cules permettent de d\u00e9finir MLBP1 comme une prot\u00e9ine liant les acyles et les lipides.<\/a><strong>&nbsp;<\/strong>met en lumi\u00e8re les interactions prot\u00e9ine-m\u00e9tabolite dans les plantes, en se concentrant sur les r\u00e9cepteurs de l'acide abscissique (ABA). Comme elle est \u00e9galement abonn\u00e9e \u00e0 mind the graph, elle a cr\u00e9\u00e9 une infographie en <a href=\"https:\/\/mindthegraph.com\">attention au graphique<\/a> pour expliquer leur m\u00e9thodologie.<\/p>\n<h2 style=\"text-align: center;\"><strong>L'importance des prot\u00e9ines li\u00e9es au domaine START<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">Selon <a href=\"https:\/\/www.nature.com\/articles\/s41598-018-22538-9\" target=\"_blank\" rel=\"noopener\">Magazine Nature<\/a>L'acide abscissique (ABA) est la principale phytohormone impliqu\u00e9e dans la r\u00e9ponse au stress abiotique et son adaptation. C'est \u00e9galement un produit agrochimique candidat.  L'ABA a \u00e9t\u00e9 d\u00e9couvert pour la premi\u00e8re fois dans les ann\u00e9es 1960. Depuis, on a d\u00e9couvert qu'il \u00e9tait un r\u00e9gulateur cl\u00e9 de processus aussi divers que la dormance, la germination, le d\u00e9veloppement des graines et les r\u00e9ponses aux stress biotiques et abiotiques.<\/p>\n<p style=\"text-align: justify;\">Les \u00e9v\u00e9nements les plus anciens impliquant la voie de signalisation de l'ABA impliquent la liaison de l'ABA aux r\u00e9cepteurs PYR\/PYL\/RCARs. Ces r\u00e9cepteurs appartiennent \u00e0 la superfamille des domaines START.<\/p>\n<p style=\"text-align: justify;\">Les domaines prot\u00e9iques START sont des domaines de liaison aux lipides pr\u00e9sents dans les g\u00e9nomes des bact\u00e9ries, des protistes, des levures, des plantes et des animaux. Cependant, ils sont plus fr\u00e9quents chez les plantes. D'apr\u00e8s<a href=\"https:\/\/genomebiology.biomedcentral.com\/articles\/10.1186\/gb-2004-5-6-r41\" target=\"_blank\" rel=\"noopener\"> Schrick<\/a>La pr\u00e9sence de domaines START chez des esp\u00e8ces \u00e9loign\u00e9es dans l'\u00e9volution, comme les animaux et les plantes, sugg\u00e8re un m\u00e9canisme conserv\u00e9 d'interaction des prot\u00e9ines avec les lipides\/st\u00e9rols.<\/p>\n<p style=\"text-align: justify;\">L'article publi\u00e9 dans <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/30735592\" target=\"_blank\" rel=\"noopener\">Le journal des plantes<\/a> se concentre sur les r\u00e9cepteurs de l'ABA PYR\/PYL\/RCARs.  Les chercheurs ont d\u00e9cid\u00e9 d'optimiser une m\u00e9thode permettant l'identification des ligands des prot\u00e9ines du domaine START et des prot\u00e9ines li\u00e9es au domaine START chez les plantes, car<em>&nbsp;\"les ligands des prot\u00e9ines du domaine START sont indispensables au bon fonctionnement de leur prot\u00e9ine tant chez les animaux que chez les plantes, et il a \u00e9t\u00e9 d\u00e9montr\u00e9 que les prot\u00e9ines du domaine START des plantes sont impliqu\u00e9es dans des processus physiologiques essentiels\".<\/em><\/p>\n<h2 style=\"text-align: center;\"><strong>Un r\u00e9sum\u00e9 visuel de la m\u00e9thodologie<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<h4 style=\"text-align: center;\"><strong>Pour expliquer leur m\u00e9thodologie, les scientifiques ont cr\u00e9\u00e9 une infographie avec des illustrations de graphiques en t\u00eate. C'est plus accrocheur et plus facile \u00e0 comprendre.&nbsp;<\/strong><\/h4>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">L'infographie ci-dessous montre le d\u00e9roulement de l'exp\u00e9rience. Comme vous pouvez le voir, ils ont exprim\u00e9 et purifi\u00e9 quatre prot\u00e9ines START. Ils ont filtr\u00e9 et incub\u00e9 le surnageant contenant la fraction soluble native \u00e0 l'aide de streptavidine s\u00e9pharose, une r\u00e9sine d'affinit\u00e9 pour une fixation fiable et \u00e9lev\u00e9e des prot\u00e9ines START.<wbr>-r\u00e9solution de la purification des biomol\u00e9cules. Puis, ils ont analys\u00e9 des aliquotes en utilisant le western blot. Pour identifier le ligand, ils ont \u00e9lu\u00e9 les ligands putatifs et les ont soumis \u00e0 une analyse par syst\u00e8me LC -MS (chromatographie liquide et spectrom\u00e9trie de masse).
