Hun er ph.d.-studerende ved fakultetet for landbrug, f\u00f8devarer og milj\u00f8 p\u00e5 Hebrew University of Jerusalem.<\/p>\n<p style=\"text-align: justify;\">Hendes nye artikel&nbsp;<a href=\"https:\/\/www.researchgate.net\/publication\/330976296_Optimized_Small-Molecule_Pull-Downs_Define_MLBP1_as_an_Acyl-Lipid-Binding_Protein\" target=\"_blank\" rel=\"noopener\">Optimerede sm\u00e5molekyle pull-downs definerer MLBP1 som et acyl-lipid-bindende protein<\/a><strong>&nbsp;<\/strong>s\u00e6tter fokus p\u00e5 protein-metabolit-interaktioner i planter, med fokus p\u00e5 abscisinsyre (ABA)-receptorer. Siden da har det vist sig at v\u00e6re en vigtig regulator af s\u00e5 forskellige processer som dvale, spiring, fr\u00f8udvikling og reaktioner p\u00e5 biotisk og abiotisk stress.<\/p>\n<p style=\"text-align: justify;\">De tidligste begivenheder, der involverer ABA-signalvejen, involverer ABA-binding til PYR\/PYL\/RCARs-receptorer. Receptorerne h\u00f8rer til START-dom\u00e6ne-superfamilien.<\/p>\n<p style=\"text-align: justify;\">START-proteindom\u00e6nerne er et lipidbindende dom\u00e6ne, der findes i genomer fra bakterier, protister, g\u00e6r, planter og dyr. De er dog mere almindelige i planter. I henhold til<a href=\"https:\/\/genomebiology.biomedcentral.com\/articles\/10.1186\/gb-2004-5-6-r41\" target=\"_blank\" rel=\"noopener\"> Schrick<\/a>Tilstedev\u00e6relsen af START-dom\u00e6ner i evolution\u00e6rt fjerne arter som dyr og planter tyder p\u00e5 en konserveret mekanisme for interaktion mellem proteiner og lipider\/steroler.<\/p>\n<p style=\"text-align: justify;\">Artiklen er udgivet i <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pubmed\/30735592\" target=\"_blank\" rel=\"noopener\">Plantejournalen<\/a> fokuserer p\u00e5 PYR\/PYL\/RCARs ABA-receptorer.  Forskerne besluttede at optimere en metode til identifikation af START-dom\u00e6ne og START-dom\u00e6ne-relaterede proteinligander i planter, fordi<em>&nbsp;\"START-dom\u00e6neproteinliganderne er uundv\u00e6rlige for, at deres protein fungerer korrekt b\u00e5de i dyr og planter, og det blev p\u00e5vist, at START-dom\u00e6neproteiner i planter er involveret i vigtige fysiologiske processer.\"<\/em><\/p>\n<h2 style=\"text-align: center;\"><strong>Et visuelt sammendrag af metoden<\/strong><\/h2>\n<p>&nbsp;<\/p>\n<h4 style=\"text-align: center;\"><strong>For at forklare deres metode har forskerne lavet en infografik med grafillustrationer. Det er i\u00f8jnefaldende og lettere at forst\u00e5.&nbsp;<\/strong><\/h4>\n<p>&nbsp;<\/p>\n<p style=\"text-align: justify;\">Infografikken nedenfor viser den eksperimentelle arbejdsgang. Som du kan se, har de udtrykt og oprenset fire START-proteiner. De filtrerede og inkuberede supernatanten, der indeholdt den native opl\u00f8selige fraktion, ved hj\u00e6lp af streptavidin-sepharose, en affinitetsresin til p\u00e5lidelige og h\u00f8je<wbr>-opl\u00f8sningsrensning af biomolekyler. Derefter analyserede de aliquoter ved hj\u00e6lp af western blot. For at identificere liganden eluerede de formodede ligander og udsatte dem for en LC -MS (v\u00e6skekromatografi og massespektrometri) systemanalyse. De tilpassede fremgangsm\u00e5den til at opdage START-ligander.
