Neoplasma af melanocytter i huden, slimhinden eller uvea er kendt som melanom. P\u00e5 trods af sin 5% repr\u00e6sentation i alle kutane maligniteter, er den ansvarlig for mere end 75% af alle hudkr\u00e6ftd\u00f8dsfald.\u00a0<\/p>\n\n\n\n<p>Selvom den positive sammenh\u00e6ng mellem UV-eksponering og udvikling af melanom er velkendt, er de underliggende epigenetiske mekanismer stort set uudforskede i humant melanomv\u00e6v. En multietnisk kohorte af 112 kutane melanomceller blev inkluderet i denne unders\u00f8gelse til klinisk, epigenom (DNA-methylom), genom- og transkriptomprofilering. MHC-I interagerer med den transport\u00f8r, der er forbundet med antigenbehandling gennem TAPBP, et medlem af immunglobulinsuperfamilien. En r\u00e6kke kr\u00e6ftformer har vist nedregulering af TAPBP-proteinet (tapasin), som gendannes efter cytokineksponering.\u00a0<\/p>\n\n\n\n<p>Det tyder p\u00e5, at manglende TAPBP-ekspression kan v\u00e6re for\u00e5rsaget af dysregulering snarere end strukturelle \u00e6ndringer. Forskerne fandt, at DNA-methyleringsniveauerne af TAPBP er signifikant omvendt relateret til transkription, og at sidstn\u00e6vnte \u00e6ndres som et resultat af UV-eksponering snarere end melanomets patologiske identitet. Hverken ikke-UV-mutante kutane eller akrale melanomer viste sig at v\u00e6re differentielt methylerede.<\/p>\n\n\n\n<p>Ud over fundene centreret om TAPBP fandt de, at UV-mutationer er forbundet med en r\u00e6kke epigenetiske \u00e6ndringer, der p\u00e5virker kutane melanomers methylom.
