ACE2 har ikke kun en enkelt struktur, det kan have flere strukturer p\u00e5 tv\u00e6rs af arter, mens det opretholder den samme funktion. <\/p>\n\n\n\n<p>Faktisk er ACE2 klassificeret som et ortologgen, dvs. et gen, der findes i forskellige arter, og som stammer fra et f\u00e6lles forf\u00e6dres gen.<\/p>\n\n\n<div class=\"wp-block-image\">\n<figure class=\"aligncenter size-large\"><a href=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes.png\"><img decoding=\"async\" loading=\"lazy\" width=\"433\" height=\"512\" src=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes.png\" alt=\"\" class=\"wp-image-12239\" srcset=\"https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes.png 433w, https:\/\/mindthegraph.com\/blog\/wp-content\/uploads\/2021\/02\/orthologs-genes-254x300.png 254w\" sizes=\"(max-width: 433px) 100vw, 433px\" \/><\/a><\/figure><\/div>\n\n\n<p>Disse strukturelle forskelle er tilsyneladende \u00e5rsagen til, at SARS-CoV-2-virussen kan binde sig til ACE2 hos nogle arter og ikke andre. <\/p>\n\n\n\n<p>N\u00e5r det virale spike-protein er forbundet med ACE2, kommer virussen ind i cellen og begynder at bruge v\u00e6rtscellens maskineri til at formere sig, hvilket f\u00f8rer til infektion.<\/p>\n\n\n\n<p>Derfor vil en bedre forst\u00e5else af ACE2-receptoren helt sikkert f\u00f8re til en mere pr\u00e6cis forklaring p\u00e5, hvorfor nogle dyrearter er modtagelige for infektioner, mens andre ikke er. <\/p>\n\n\n\n<p>Derudover vil det ogs\u00e5 hj\u00e6lpe forskere med at udvikle l\u00e6gemidler ved at anvende antivirale strategier, som f.eks. at producere falske skabe, som virussen kan binde sig til i stedet for v\u00e6rtens egentlige skab. <\/p>\n\n\n\n<p>Samt for bedre at kunne spore mellemv\u00e6rtsdyr for fremtidige virusinfektioner.&nbsp;<\/p>\n\n\n\n<p>M\u00e5let med projektet var derfor at karakterisere den tredimensionelle struktur af ACE2 og dens interaktioner p\u00e5 atomart niveau ved hj\u00e6lp af computermodellering.<a href=\"https:\/\/journals.plos.org\/ploscompbiol\/article?id=10.1371\/journal.pcbi.1008449\"> Rodrigues og kolleger<\/a> arbejde. <\/p>\n\n\n\n<p>Denne type karakteriserende unders\u00f8gelse er eksperimentelt dyr og ikke anvendelig i stor skala, hvilket g\u00f8r det sv\u00e6rt at n\u00e5 frem til afg\u00f8rende data, mens beregningsmodellering er hurtig og billig og meget gavnlig til at udfylde nogle huller i viden om strukturelle rammer.<\/p>\n\n\n\n<p class=\"has-large-font-size\">Unders\u00f8gelsen tog udgangspunkt i 28 dyrearter og skabte en 3D-model af ACE2 bundet til det virale spike-protein. <\/p>\n\n\n\n<p>Resultaterne viste, at der er vigtige steder i ACE2-aminosyresekvensen, hvor virusproteinet p\u00e5 grund af mutationer ikke kunne binde sig til v\u00e6rtscellen. <\/p>\n\n\n\n<p>Specifikt mangler ACE2-sekvensen en aminosyre, der er ansvarlig for en intermolekyl\u00e6r saltbro mellem ACE2 og bindingspunktet for det virale protein, som rapporteret af Rodrigues og hans team. <\/p>\n\n\n\n<p>Dette resultat underst\u00f8ttes af andre offentliggjorte studier, som eksperimentelt har vist, at denne mutant mangler en negativt ladet aminosyre, som g\u00f8r det muligt for virusproteinet at binde sig.<\/p>\n\n\n\n<p class=\"has-large-font-size\">Unders\u00f8gelsen indikerer ogs\u00e5, at SARS-CoV-2-infektionen er mere kompleks end tidligere antaget, med mere end \u00e9n m\u00e5de at inficere v\u00e6rten p\u00e5. <\/p>\n\n\n\n<p>Det er derfor vigtigt at pr\u00e6cisere, at en reduktion af ACE2-tilg\u00e6ngeligheden kan mindske risikoen for infektion, men det forhindrer ikke, at infektionen opst\u00e5r.<\/p>\n\n\n\n<p>Og at beregningsmodellerne ikke kan bruges alene til at forudsige arternes risiko for infektion - yderligere eksperimentelle data er n\u00f8dvendige for at f\u00e5 en mere p\u00e5lidelig konklusion.<\/p>\n\n\n\n<p>S\u00e5 jeg tror, vi skal forts\u00e6tte med at f\u00f8lge pandemiprotokollerne og g\u00f8re, hvad vi allerede har gjort ...
