Naivn\u00ed net\u0159\u00edd\u011bn\u00e9 bu\u0148ky byly kultivov\u00e1ny v CSC-mediu obsahuj\u00edc\u00edm FGF2 nebo s inhibitorem FGFR SU-5402.<\/p>\n\n\n\n<h2><strong>Byla zkoum\u00e1na role fibroblastov\u00e9ho r\u016fstov\u00e9ho faktoru 2 (FGF2) v sebeobnov\u011b kmenov\u00fdch bun\u011bk kolorekt\u00e1ln\u00edho karcinomu.<\/strong><\/h2>\n\n\n\n<p>V \u010dl\u00e1nku \"FGF Signalling in the Self-Renewal of Colon Cancer Organoids\" J\u00f6rg Otte a jeho kolegov\u00e9 (2019) uv\u00e1d\u011bj\u00ed, \u017ee kdy\u017e byly naivn\u00ed jednotliv\u00e9 bu\u0148ky vlo\u017eeny do Matrigelu a kultivov\u00e1ny v CSC-mediu, byla u v\u0161ech analyzovan\u00fdch pacient\u016f pozorov\u00e1na komplexn\u011bj\u0161\u00ed sebeorganizace. <\/p>\n\n\n\n<p>Bylo zji\u0161t\u011bno, \u017ee geny p\u0159\u00edmo spojen\u00e9 s EMT spolu s mezenchym\u00e1ln\u00edm a sebeobnovuj\u00edc\u00edm fenotypem (TGFB3, ID1, ID2 a ID4) jsou v organoidech nadm\u011brn\u011b exprimov\u00e1ny. <\/p>\n\n\n\n<p>Zat\u00edmco inhibice FGFR vedla k pos\u00edlen\u00ed signalizace MAPK, pozorovali indukovanou diferenciaci, ztr\u00e1tu v\u011bt\u0161iny marker\u016f kmenov\u00fdch bun\u011bk a epiteli\u00e1ln\u00ed fenotyp. <\/p>\n\n\n\n<p>N\u00e1dorov\u00e9 kmenov\u00e9 bu\u0148ky jsou subpopulac\u00ed malign\u00edch bun\u011bk schopn\u00fdch sebeobnovy a slou\u017e\u00edc\u00edch jako trval\u00fd zdroj diferencovan\u00fdch n\u00e1dorov\u00fdch bun\u011bk.
